We report a method to detect proteins via suppression of rolling circle amplification (RCA) by using an appropriate aptamer as the linear primer (denoted as an aptaprimer) to initiate RCA. In the absence of a protein target, the aptaprimer is free to initiate RCA, which can produce long DNA products that are detected via binding of a fluorescent intercalating dye. Introduction of a target causes the primer region within the aptamer to become unavailable for binding to the circular template, inhibiting RCA. Using SYBR Gold or QuantiFluor dyes as fluorescent probes to bind to the RCA reaction product, it is possible to produce a generic protein-modulated RCA assay system that does not require fluorophore- or biotin-modified DNA species, substantially reducing complexity and cost of reagents. Based on this modulation of RCA, we demonstrate the ability to produce both solution and paper-based assays for rapid and quantitative detection of proteins including platelet derived growth factor and thrombin. 相似文献

N型隧穿氧化层钝化接触(Tunnel Oxide Passivating Contacts,TOPCon)太阳能电池完成印刷烧结后,再经过光注入,效率有明显提升,主要表现在Voc(开路电压)及FF(填充因子)的提升。其机理在于通过温度和光照强度调节费米能级变化,控制H总量及价态来提高钝化性能。钝化膜层的质量、硅基体掺杂浓度、光注入退火时的工艺温度等对光注入退火工艺提升效率有很大影响。实验证明转换效率越低的电池片经过光注入后效率提升幅度越大;转换效率越高的电池片缺陷会更小,经过光注入退火工艺后几乎无增益。另外同心圆在经过光注入退火工艺后会明显消除。本文主要研究温度、光强、基体电阻率、正表面金属接触面积大小、poly-Si(多晶硅)厚度对光注入退火工艺增效的影响。 相似文献

The relationship between the number of summable solutions of non-self-adjoint difference equations and that of the discrete linear Hamiltonian system is discussed. In the present study, an inductively-coupled plasma-mass spectrometry (ICP-MS)-based triple-amplification system, by combination of nicking–displacement, rolling circle amplification (RCA) and bio-bar-code probes, was fabricated for the detection of DNA target. By using this system, hepatitis B virus (HBV) DNA target down to 3.2 × 10−17 M was detected by DNA probes labeled with Au nanoparticles (AuNPs). Single nucleotide polymorphisms in genes can also be effectively discriminated. In addition, we proved that this strategy is capable of detecting the target in complicated biological samples and holds great potential application in biomedical research. 相似文献

An analytical solution is presented for the calculation of the flow field in a concentric cylinder viscometer of non-ideal Bingham-fluids, described by the Worrall-Tuliani rheological model. The obtained shear rate distribution is a function of the a priori unknown rheological parameters. It is shown that by applying an iterative procedure experimental data can be processed in order to obtain the proper shear rate correction and the four rheological parameters of the Worrall-Tuliani model as well as the yield surface radius. A comparison with Krieger's correction method is made. Rheometrical data for dense cohesive sediment suspensions have been reviewed in the light of this new method. For these suspensions velocity profiles over the gap are computed and the shear layer thicknesses were found to be comparable to visual observations. It can be concluded that at low rotation speeds the actually sheared layer is too narrow to fullfill the gap width requirement for granular suspensions and slip appears to be unavoidable, even when the material is sheared within itself. The only way to obtain meaningfull measurements in a concentric cylinder viscometer at low shear rates seems to be by increasing the radii of the viscometer.
